Cre-loxP Systems in Drosophila Transgenesis

The Cre-loxP system is a powerful tool used in Drosophila transgenesis to achieve precise genetic modifications. It consists of two main components: the Cre recombinase enzyme and the loxP DNA recognition sites. The Cre recombinase recognizes and binds to specific loxP sites, which are short DNA sequences. When Cre is present, it catalyzes a recombination event between two loxP sites, resulting in the excision, inversion, or translocation of DNA sequences located between these sites.

In Drosophila transgenesis, loxP sites are strategically placed within the genome, flanking the target gene or regulatory elements of interest. By introducing a transgene expressing Cre recombinase into the flies, researchers can induce recombination between the loxP sites, leading to the desired genetic modification. This can include gene knockout, gene activation, or even tissue-specific gene manipulation. The Cre-loxP system offers precise control and flexibility, allowing for the investigation of gene function and regulation in a spatiotemporal manner, making it an invaluable tool in Drosophila research.

Our Services

We specialize in providing cutting-edge services in the field of Drosophila research. Our customized transgenesis services based on the Cre-loxP system offer precise genetic manipulation and targeted gene expression, enabling researchers to advance their studies with unparalleled control and specificity. With our expertise and state-of-the-art techniques, we assist researchers in generating transgenic Drosophila lines tailored to their specific needs. Here are the key components of our service:

Workflow.

Consultation and Project Design

Our team of experts will work with you to design a customized transgenesis strategy based on the Cre-loxP system.

We take into account factors such as genes of interest, desired genetic modifications, and specific experimental conditions to ensure optimal results.

Construct Design and Generation

Once the project design is finalized, our team will proceed with designing the necessary DNA constructs for your transgenesis experiment.

This includes creating the loxP-flanked target construct, as well as the Cre recombinase expression construct, tailored to your specific needs.

Transgenesis and Embryo Injection

Performing the actual transgenesis process, introducing the designed constructs into Drosophila embryos using established techniques. This step may involve microinjection or other suitable methods, depending on the requirements of your project.

Cre-loxP Recombination and Genetic Modification

Once transgenic lines are established, we initiate the Cre-loxP recombination process to achieve precise genetic modifications. By activating the Cre recombinase, we can excise or rearrange DNA sequences flanked by loxP sites.

This enables the specific manipulation of genes or regulatory elements, facilitating the study of gene function and regulation in Drosophila models.

Characterization and Phenotypic Analysis

Conducting comprehensive characterization and phenotypic analysis of the resulting transgenic lines. This may include molecular assays, such as genome sequencing, as well as phenotypic assessments to evaluate the effects of the genetic modifications on the Drosophila model.

Data Analysis and Reporting

Upon completion of the experimental work, we provide detailed reports summarizing the experimental procedures, results, and findings.
